Research Description

My research focuses on a crucial cellular process: meiosis. Meiosis, the specialized form of cell division responsible for generating sperm and oocytes, is a cornerstone of reproductive biology. Employing cutting-edge micro-manipulation techniques, state-of-the-art sequencing methodologies, and advanced computational tools, we have pioneered new avenues of exploration within the realm of meiosis. Through these sophisticated tools, we have meticulously dissected meiotic checkpoints, elucidated intricate chromosome structures, and discerned the impact of environmental factors on meiotic processes. Moreover, we have developed novel computational approaches to dissect the transcriptional and 4D genomic landscapes of meiotic chromosomes. Our live-seq allows us to link the transcriptomic changes with individual cell phenotypes. Additionally, our innovative "digital chromosome banding" method, based on single-cell RNA-seq, enables the precise detection of transcriptionally silenced chromosome regions. Embracing a multidisciplinary approach, we engineer oocytes alongside their spindles and chromosomes, effectively utilizing oocytes as a versatile "culture dish" and “reaction chamber” to probe myriad biophysical and molecular questions of interest. Our work represents a convergence of cutting-edge techniques and theoretical frameworks, driving forward our understanding of meiosis and its implications in reproductive biology.
